DNA methylation is the most widely studied epigenetic modification. DNA methylation occurs mainly on cytosine bases in the CpG dinucleotide motif in mammalian genomes, and across all cytosine contexts in plant genomes. DNA methylation is often associated with the regulation of gene expression through its presence or absence in CpG Islands among gene promoter regions, and these methylation patterns can be used to identify different cell types.  

Swift Accel-NGS Methyl-Seq DNA Library Kit

The Accel-NGS Methyl-Seq DNA Library Prep utilizes Adaptase® technology for capturing single-stranded DNA molecules in an unbiased manner. As bisulfite treatment of samples damages DNA, Swift Biosciences’ single-strand compatible Adaptase technology maximizes recovery of DNA from bisulfite-converted samples. The resultant libraries accurately represent sample base composition of the entire genome for the analyzed organism.

The technology has quickly become the gold standard for providing the most comprehensive coverage of the methylome in a simple, 2-hour protocol. It is an excellent choice for whole genome bisulfite sequencing as well as targeted bisulfite sequencing applications, such as RRBS and hybridization capture using the NimbleGen™ SeqCap™ Epi Enrichment Sysem.

The Accel-NGS Methyl-Seq Kit is also compatible with bisulfite-converted DNA samples enriched by ChIP or other methods. This library preparation technology contains a uracil tolerant polymerase which is ideal for sequencing ancient DNA samples that may contain uracil nucleotides as a result of damage. A separate Accel-NGS Adaptase Module for High Throughput Single-Cell Methyl-Seq is available.

The EpiNext™ Post-Bisulfite DNA Library Preparation Kit (Illumina) is a complete set of optimized reagents to prepare a DNA library after successful bisulfite conversion. The EpiNext™ High-Sensitivity Bisulfite-Seq Kit is designed to bisulfite-convert DNA and prepare an Illumina-based library for bisulfite sequencing, all in one kit. The optimized protocol and components of the kits allow subnanogram amounts of DNA to be bisulfite converted and fragmented simultaneously followed by quick non-barcoded (singleplexed) and barcoded (multiplexed) library construction in less than 7 hours.
